Examine the drop - U-tube method

Introduction

In this method for detection of bacterial motility ‘U’ shaped tube is used. The tube is filled with semi solid agar. The bacteria are inoculated at one end of the tube. The motile bacteria will come on to the other end of the tube while the non-motile bacteria will remain at the site of inoculation.

Requirements

  1. Bacterial culture
  2. U-tube filled with semi solid agar
  3. Inoculating loop (straight wire)
  4. Bunsen burner
  5. Equipments
  6. Incubator

Procedure

  1. Inoculate the 24 hrs bacterial culture into one arm of the U-tube.
  2. Incubate the U-tube in upright position overnight.
  3. Next day observe the tube for growth of bacteria on the other end of the U-tube.
  4. Interpret the results in the following manner.
  5. Growth on the other end – motile bacteria
  6. No growth on the other end – non-motile bacterial
